No. A stock gain can increase your tax, but it does not automatically mean you should send an estimated payment.
First compare your withholding and prior payments with the federal safe-harbor rules. The timing of the gain can also matter, and a W-2 employee may sometimes increase wage withholding instead of making a separate estimated payment.
